Approaching ‘ kit - type ’ labelling with 68 Ga : The the DATA chelators
نویسندگان
چکیده
The DATA chelators, are a novel class of tri-anionic chelators ligands based on the 6-amino1,4-diazepine-triacetic acid, core structure, that have been introduced recently for the chelation of Ga. Compared to established macrocyclic chelators based on the cyclen scaffold (i.e. DOTA-, DO3Aand DO2Aderivatives), they enable undergo quantitative radiolabelling both more rapidly and under considerably milder conditions. To date, a detailed examination of the radiochemical characteristics of these chelators had not been carried out. In this study, we present a systematic evaluation of the labelling of four DATA chelators – DATA, DATA, DATA and DATA. – by Ga is presented The results of this study underlinehighlight the extraordinary potential of this novel new class of chelators for application in molecular imaging using Ga-PETpositron emission tomography (PET).

DATATOC: a novel conjugate for kit-type 68Ga labelling of TOC at ambient temperature
Background The widespread acceptance and application of 68Ga-PET depends on our ability to develop radiopharmaceuticals that can be prepared in a convenient and suitable manner. A kit-type labelling protocol provides such characteristics and requires chelators that can be radiolabelled under exceptionally mild conditions. Recently the DATA chelators have been introduced that fulfil these requir...
متن کاملRapid kit-based 68Ga-labelling and PET imaging with THP-Tyr3-octreotate: a preliminary comparison with DOTA-Tyr3-octreotate
BACKGROUND Ge/(68)Ga generators provide an inexpensive source of a PET isotope to hospitals without cyclotron facilities. The development of new (68)Ga-based molecular imaging agents and subsequent clinical translation would be greatly facilitated by simplification of radiochemical syntheses. We report the properties of a tris(hydroxypyridinone) conjugate of the SSTR2-targeted peptide, Tyr(3)-o...
متن کاملComparison of macrocyclic and acyclic chelators for gallium-68 radiolabelling.
Gallium-68 (68Ga) is a positron-emitting isotope used for clinical PET imaging of peptide receptor expression. 68Ga radiopharmaceuticals used in molecular PET imaging consist of disease-targeting biomolecules tethered to chelators that complex 68Ga3+. Ideally, the chelator will rapidly, quantitatively and stably coordinate 68Ga3+ at room temperature, near neutral pH and low chelator concentrati...
متن کاملInfluence of Macrocyclic Chelators on the Targeting Properties of 68Ga-Labeled Synthetic Affibody Molecules: Comparison with 111In-Labeled Counterparts
Affibody molecules are a class of small (7 kDa) non-immunoglobulin scaffold-based affinity proteins, which have demonstrated substantial potential as probes for radionuclide molecular imaging. The use of positron emission tomography (PET) would further increase the resolution and quantification accuracy of Affibody-based imaging. The rapid in vivo kinetics of Affibody molecules permit the use o...
متن کاملNew Tris(hydroxypyridinone) Bifunctional Chelators Containing Isothiocyanate Groups Provide a Versatile Platform for Rapid One-Step Labeling and PET Imaging with 68Ga3+
Two new bifunctional tris(hydroxypyridinone) (THP) chelators designed specifically for rapid labeling with (68)Ga have been synthesized, each with pendant isothiocyanate groups and three 1,6-dimethyl-3-hydroxypyridin-4-one groups. Both compounds have been conjugated with the primary amine group of a cyclic integrin targeting peptide, RGD. Each conjugate can be radiolabeled and formulated by tre...
